PMEA transport Cells were seeded at a density of 2 x 106 per well in poly-D-lysine-coated 12-well plates and grown overnight. The next day the cells were washed with PBS and loaded for 1 hour with the PMEA pro-drug bis-POM-[3H]PMEA 1 M ; in pre-warmed ATP-depletion medium DMEM without glucose Life Technologies ; supplemented with 10 mM sodium azide and 10 mM 2-deoxyglucose. After loading, the cells were washed quickly with pre-warmed PBS and incubated at 37 C pre-warmed HBSS supplemented with the compounds indicated in the text.
