Another limitation of this study was the comparison of vascular smooth muscles from different vascular beds and different species. Nevertheless, we show that transduced bioactive molecules can attenuate smooth muscle contraction. An advantage of this system was that immunoblots of tissue homogenates could be performed to correlate the relative amounts of intracellular HSP20 with physiological responses. In addition, this physiological activity was dependent on a phosphorylated bioactive protein. Finally, this approach bypasses receptors, signaling molecules, and gene expression and allows for a functional assessment of the activity of a protein in human tissue. Cultured oligodendrocytes grown on polyLornithine-coated coverslips were loaded with 10 M fura-2-AM in HEPES-MEM at room temperature for 30 min. The coverslips were placed in the open bath imaging chamber containing HEPES-MEM at room temperature. The chamber was mounted on the stage of the Nikon TE 300 inverted epifluorescence microscope. Oligodendrocytes were excited every 10 s at 340 and 380 nm and the emission fluorescence at 510 nm was recorded. The origins of April Fool's Day are unknown, although various theories have been proposed. It is considered to be related to the festival of the vernal equinox, which occurs on March 21. Prior to the adoption of the Gregorian calendar in 1582, it was observed as New Year's Day by cultures as far apart as ancient Rome and India. New Year was originally celebrated from March 25 to April 1, before the Gregorian reforms moved it back to January 1. The English first celebrated the day on a widespread basis only as late as the 18th century, though it appears to have reached England probably from Germany in the mid-17th century. Its first known description in English originates with John Aubrey, who noted in 1686: "Fooles holy day. We observe it on the first of April. And so it is kept in Germany everywhere." The custom of playing practical jokes on April Fool's Day is also very widespread and of uncertain origins. The victim of a joke is known in English as an April Fool; in Scots as a "gowk" cuckoo or fool and in French as a poisson d'avril April fish ; . Pentamidine dosage
Suggest that particular classes of molecules interact preferentially with different targets within the parasite. Although pentamidine is used clinically for the treatment of leishmaniasis, this compound has very poor activity in antileishmanial assays measuring the effects of drug on parasiteinfected macrophages 23 ; . Agents with potency comparable or superior to that of pentamidine in the axenic amastigote assay were examined in our Leishmania-infected macrophage model in an attempt to identify candidate compounds for further study in vivo. Unfortunately, we did not observe activity in our infected macrophage cell line with any of the compounds described in this paper. We are optimistic, however, about the potential of related dicationic molecules as antileishmanial drug candidates. Information obtained from the structure-activity relationships outlined here is currently being used to design new molecules that have displayed superior antileishmanial activity in preliminary experiments, and we look forward to providing future accounts of the antiparasitic potential of novel aromatic dications and pergolide.

The potent physiologic effects of PGs and TBx and their possible role in RSV infection suggest that these mediators should be considered as potential important factors in the RSV disease process. The animal model of choice for RSV studies is the cotton rat Sigmodon hispidus ; , because it most closely resembles disease in humans 19 ; . Preclinical data from cotton rats have resulted in a successful prophylactic approach in humans 20 24 ; . this work we present data demonstrating the induction of COX-2 expression during RSV infection in vitro, in human lung alveolar epithelial cells and cotton rat macrophages, and in vivo, in the lungs of cotton rats infected with RSV.
